(S)-(-)-4-羟基-2-吡咯烷酮性质、用途与生产工艺

用途

(S)-(-)-4-羟基-2-吡咯烷酮是(S)-奥拉西坦的中间体。奥拉西坦临床上用于轻中度血管性痴呆、老年性痴呆以及脑外伤等症引起的记忆与智能障碍，该化合物1974年由意大利史克比切姆公司合成，该药是吡拉西坦的β-羟基衍生物，是一种羟基氨基丁酸(GABOB)的环状衍生物。

制备

将2-环氧乙烷基乙酰胺10.1g(100mmol)、CuI 0.95g(5mmol)、碳酸铯2g(200mmol)加入到150ml氨气的甲醇饱和溶液中，55℃进行搅拌反应8h，反应液减压浓缩，100ml加水，二氯甲烷萃取三次，饱和食盐水洗涤三次，乙酸乙酯相减压浓缩，石油醚重结晶得到(S)-(-)-4-羟基-2-吡咯烷酮8.71g，收率86.3％，HPLC纯度99.42％(面积归一法)，ee值99.67％。
