磷酸烯醇式丙酮酸羧化酶性质、用途与生产工艺
磷酸烯醇丙酮酸羧化激酶是以一条单一肽链构成,相对分子质量为740千道尔顿的一种激酶。在磷酸烯醇丙酮酸羧化激酶的催化下,草酰乙酸被转变为磷酸烯醇式丙酮酸和二氧化碳。该反应消耗一分子的鸟苷三磷酸以提供磷酰基。在糖异生作用中,此酶与丙酮酸羧化酶一起构成了从丙酮酸转化为磷酸烯醇丙酮酸的迂回步骤。
磷酸烯醇式丙酮酸羧化酶在植物和细菌中广泛存在,在动物及丝状霉菌中缺乏此酶。在植物的叶绿体和细菌的可溶性分级沉淀中存在。
磷酸烯醇式丙酮酸羧化酶(PEPC)催化磷酸烯醇式丙酮酸(PEP)和HCO3-生成草酰乙酸(OAA)和无机磷酸的不可逆反应。
磷酸烯醇式丙酮酸羧化酶是植物细胞代谢中的一种重要羧化酶,在不同类型植物中执行着不同的功能。在C4和景天酸代谢(CAM)植物中,它起着捕获CO2及作为CO2泵以提高光合效率的功能,该酶介导不可逆的β羧化反应,将磷酸烯醇式丙酮酸转化成草酰乙酸和磷酸。而在C3植物中,该酶为Krebs循环补充四碳二羧酸水平而起着回补反应的功能。
