Uses
5-(Benzyloxy)resorcinol is an intermediate used in the synthesis of Phloroglucinol Glucuronide (P339995), which is used in preliminary clinical studies with ibenzmethyzin for hodgkin’s disease and melanoma treatment.
Synthesis
Phloroglucinol, (3 g, 23 mmol) was dissolved in anhydrous DMF (10 mL) and added to suspension of K2CO3 (1.44 g, 10 mmol) in DMF (10 mL). The mixture was stirred for 10 minutes. Benzyl bromide (1.76 g, 1 mmol) in DMF (5 mL) was added to mixture dropwise over 40 minutes. The brown colored mixture was stirred 1 more hour and inorganic salts were removed by filteration. DMF was evaporated and the crude mixture was dissolved in EtOAc and washed with 1N HCl solution (3x20 mL). Organic layers was combined and purified by column chromatography on silica gel with THF- CH2Cl2 (7:93) to give a light brown oil in 40% yield. 1 H NMR (250 MHz, DMSO-d6) δ 9.1(2H), 7.3(5H), 5.8(3H), 4.9 (2H). GC-MS Cald C13H12O3 + : 216.0; found: 216.2
