以1H-吡唑并[3,4-c]吡啶为原料合成3-碘-1H-吡唑并[3,4-c]吡啶的一般步骤:将1H-吡唑并[3,4-c]吡啶(300 mg,2.52 mmol,1.00当量)、氢氧化钾(KOH,500 mg,8.91 mmol,3.50当量)和二碘甲烷(1.28 g,5.04 mmol,2.00当量)悬浮于N,N-二甲基甲酰胺(DMF,10 mL)中。反应混合物在室温下搅拌过夜。反应完成后,用水淬灭反应,用乙酸乙酯萃取有机相,有机层用无水硫酸钠干燥,减压浓缩。粗产物通过硅胶柱色谱纯化,洗脱剂为乙酸乙酯/石油醚(1:1,v/v),得到3-碘-1H-吡唑并[3,4-c]吡啶(577 mg,收率84%)为黄色固体。LC-MS(ESI,m/z):246 [M + H]+。
