以2-氧代-1,2-二氢-3-吡啶羧酸甲酯为原料合成5-碘-2-氧代-1,2-二氢吡啶-3-甲酸甲酯的一般步骤如下:在室温条件下,将2-羟基烟酸甲酯(20g)溶解于二氯甲烷(500mL)中,随后加入N-碘代琥珀酰亚胺(NIS,38g)。将反应混合物加热至回流状态,持续反应16小时。反应完成后,蒸发去除溶剂。向残余物中加入乙酸乙酯(200mL),并在回流条件下加热2小时。反应混合物冷却后,过滤收集不溶性固体,得到目标产物2-羟基-5-碘烟酸甲酯(30g,收率81%),为白色固体。产物经1H NMR(300MHz,CDCl3)表征:δ3.97(s,3H),8.33(brs,1H),8.43(d,J=2.4Hz,1H)。
