c)4-氯-1-(二氟甲基)-1H-吡唑-3-羧酸的合成:将4-氯-1-(二氟甲基)-1H-吡唑-3-甲酸甲酯(540 mg,2.6 mmol)溶于四氢呋喃(18 mL)中,室温下缓慢加入氢氧化锂(135 mg,5.6 mmol)的水和甲醇(1:1,12 mL)混合溶液。反应1小时后,减压浓缩除去大部分溶剂。残余物溶于水(10 mL)中,用2M盐酸酸化至pH≈2。随后用乙酸乙酯萃取,有机相用无水硫酸钠干燥,减压浓缩得到白色固体粗产物(555 mg)。将粗产物用戊烷(10 mL)研磨,过滤收集固体,并用戊烷洗涤,减压干燥后得到4-氯-1-(二氟甲基)-1H-吡唑-3-羧酸(477 mg,收率95%)。质谱(ISP):m/z = 195.0 [M-H]?。
