步骤2)合成2,4-二氯-6-氟喹唑啉:向三氯氧化磷(46.0 mL,502.5 mmol)中加入五氯化磷(12.5 g,60.0 mmol),随后在搅拌下缓慢加入6-氟喹唑啉-2,4(1H,3H)-二酮(3.6 g,20.0 mmol)。将反应混合物加热至回流并搅拌9小时。反应完成后,冷却混合物并在真空下除去溶剂。向残余物中加入冰水混合物(400 mL),继续搅拌0.5小时。用二氯甲烷(DCM,250 mL×3)萃取混合物。合并DCM层,用无水硫酸钠干燥,过滤,并将滤液在真空下浓缩。通过硅胶柱色谱法(洗脱剂:石油醚/乙酸乙酯,v/v=30/1)纯化残余物,得到标题化合物2,4-二氯-6-氟喹唑啉,为白色固体(3.735 g,收率86.0%)。MS(ESI,阳离子)m/z:216.9 [M + H]+;1H NMR(CDCl3,400 MHz)δ(ppm):8.03(dd,J=9.2 Hz,4.9 Hz,1H),7.86(dd,J=8.1 Hz,2.7 Hz,1H),7.79-7.73(m,1H)。
