在氮气气氛下,将乙酸钯(II)(0.084 g,0.38 mmol)与xantphos(0.220 g,0.38 mmol)在25 mL四氢呋喃中混合。搅拌5分钟后,将所得溶液转移至含有2,4-二溴噻唑(3.64 g,15 mmol)、苯硼酸(1.96 g,16 mmol)和磷酸钾(9.55 g,45 mmol)的圆底烧瓶中。将反应混合物在60℃下加热18小时,随后冷却至室温,过滤并用二氯甲烷洗涤。在减压下除去溶剂,得到粗产物,通过硅胶柱色谱法纯化,使用己烷:乙酸乙酯作为洗脱剂,得到4-溴-2-苯基噻唑,为白色固体(3 g,产率84%)。1H NMR(400 MHz,CDCl3)δ: 7.95(dd,J = 3.4 Hz,8.2 Hz,2H),7.47-7.46(m,3H),7.23(s,1H)。LRMS(ESI)计算值C9H6BrNS [M + H]+: 241.12,实测值: 241.00。
