步骤3:4,6-二氯-5-硝基烟酸乙酯(16)的制备 将4,6-二羟基-5-硝基烟酸乙酯(15)(20.0 g,87.7 mmol)溶于磷酰氯(80 mL,874 mmol)中。将反应混合物在80℃下搅拌24小时。反应完成后,在减压条件下蒸除约一半体积的磷酰氯。将残余物缓慢倒入冰水中,并用乙酸乙酯萃取两次。合并有机相,依次用碳酸钠水溶液、水和饱和食盐水洗涤。有机层用无水硫酸钠干燥,过滤后减压浓缩,得到棕色油状粗产物。粗产物通过硅胶柱色谱法纯化,以石油醚/乙酸乙酯(3:1,v/v)为洗脱剂,得到白色结晶固体16(14.2 g,产率61.1%)。产物熔点为36-38℃。1H NMR(300 MHz,DMSO-d6)δ:9.07(s,1H),4.39(q,J = 6.0 Hz,2H),1.34(t,J = 6.0 Hz,3H)。MS(ESI(+)70V)m/z:264 [M + H]+。
