以5-三甲基硅乙炔基-2,2'-联吡啶为原料合成5-炔基-2,2'-联吡啶的一般步骤:将5-[(三甲基甲硅烷基)乙炔基]-2,2'-联吡啶(0.360 g,1.43 mmol)溶于30 mL THF(预先用N2脱气)。随后,加入KOH(0.320 g,5.72 mmol)的甲醇(30 mL)溶液。反应混合物在室温下搅拌6小时。反应完成后,通过减压蒸馏除去溶剂。残余物通过快速硅胶柱色谱法纯化,使用二氯甲烷作为洗脱剂。产物收率为95%(245 mg)。薄层色谱(TLC,SiO2)显示Rf值为0.45(10%甲醇/二氯甲烷)。红外光谱(IR,KBr,cm-1)显示特征吸收峰:3298(C≡CH),3190(C≡CH),3049(Ar-H),3001(Ar-H),2964(Ar-H),2933(Ar-H),2096(C≡C)。1H NMR(400 MHz,CDCl3)δ 3.29(s,1H,C≡CH),7.26-7.33(m,1H,PyH),7.79-7.84(m,1H,PyH),7.88-7.90(m,1H,PyH),8.37-8.41(m,2H,PyH),8.67-8.69(m,1H,PyH),8.77(m,1H,PyH)。质谱(MS/EI)显示m/z 180 [M+,100%],153 [M+-NCH,48%],126 [M+-2(NCH),9%]。
