以2-甲氧基-4-氟苯腈为原料合成4-氟-2-羟基苯腈的一般步骤:将4-氟-2-甲氧基苄腈(4.53 g,30 mmol)与三氯化铝(5.0 g,37.6 mmol;购自Aldrich)在无水甲苯(30 mL)中的混合物于130℃下搅拌反应18小时。反应完成后,将混合物冷却至室温,加入冰水(约50 mL),用乙醚(20 mL × 2)萃取。合并有机相,依次用水和饱和盐水洗涤,然后用无水硫酸镁干燥。减压浓缩后,得到4-氟-2-羟基苯腈(3.90 g,28.5 mmol,收率95%),为白色固体。其结构通过以下表征确认:1H NMR(DMSO-d6, 300 MHz)δ ppm:6.74-6.84(2H,m,Ar-H),7.71(1H,dd,J = 7 Hz,J = 8.5 Hz,Ar-H),11.64(1H,s,OH);13C NMR(DMSO-d6, 75.5 MHz)δ ppm:95.13, 102.45, 102.78, 106.53, 106.83, 115.53, 134.68, 134.84, 161.41, 161.58, 163.00, 166.35。高分辨质谱(ESI-)计算值C7H3NOF(M-H)为136.0199,实测值为136.0199。
