步骤1:以6-氯吡啶-3-醇(112g,868mmol)为原料,将其溶解于THF(800mL)和水(800mL)的混合溶剂中。随后加入碳酸钠(92.0g,1.736mol)和碘(244.2g,1.04mol)。将反应混合物在室温下搅拌4小时。反应完成后,分离水相和有机相,产物主要存在于水相中。用水相产物经己烷(400mL×2)洗涤以去除杂质。接着,用盐酸将水相的pH值调节至7,然后用乙酸乙酯(500mL×4)进行萃取。合并有机层,用无水硫酸钠干燥,过滤。滤液经减压浓缩得到粗产物,粗产物用乙酸乙酯洗涤,最终得到6-氯-2-碘吡啶-3-醇163g,产率为75%。产物结构经1H NMR(DMSO,400MHz)确认:δ11.13(s,1H,OH),7.31(dd,J = 8.4,12.8Hz,2H)。
