步骤1:在-78℃条件下,将6-甲氧基-3,4-二氢异喹啉-1(2H)-酮(2.58g,14mmol)溶于二氯甲烷中,缓慢加入三溴化硼(2.7mL,28mmol)。反应混合物逐渐升温至室温并搅拌过夜。反应完成后,用冷水淬灭反应,随后用乙酸乙酯萃取。合并有机相,真空浓缩去除溶剂。粗产物通过ISCO CombiFlash?系统进行纯化,采用硅胶柱色谱法,以0-15%甲醇的二氯甲烷溶液为洗脱剂,得到6-羟基-3,4-二氢异喹啉-1-酮,为浅棕色固体(1.8g,收率75%),熔点为204-206℃;质谱(电喷雾离子化)m/z 162.1 [M+H]+。
