8-甲基-1,4-二氧杂螺[4.5]癸烷-8-羧酸乙酯的合成步骤如下:在0℃下,向二异丙胺(DIA,1.2当量)的无水THF(1M)溶液中缓慢滴加正丁基锂(n-BuLi,1.6当量),历时20分钟。将反应混合物在0℃下搅拌30分钟后,进一步冷却至-78℃。随后,缓慢滴加1,4-二氧杂螺[4.5]癸烷-8-羧酸乙酯(1当量)的THF(1M)溶液,历时30分钟,然后加入碘甲烷(3当量)。将反应混合物在室温下搅拌过夜。反应完成后,用盐水稀释反应混合物,并用乙酸乙酯萃取。合并有机层,用盐水洗涤,无水硫酸钠(Na2SO4)干燥,过滤。浓缩滤液,得到目标产物8-甲基-1,4-二氧杂螺[4.5]癸烷-8-羧酸乙酯,收率100%。产物经1H NMR(400MHz,CDCl3)表征,化学位移δ(ppm)如下:4.16(q,J=7.1Hz,2H),3.95(s,4H),2.19-2.11(m,2H),1.72-1.57(m,4H),1.57-1.45(m,2H),1.30-1.24(m,3H),1.20(s,3H)。
