I. 合成N-(6-溴吡啶-2-基)乙酰胺的一般步骤: 步骤a:在0℃条件下,向6-溴吡啶-2-胺(10g,0.060mol)和三乙胺(Et3N,25g,0.27mol)的二氯甲烷(CH2Cl2,300mL)溶液中缓慢加入乙酰氯(AcCl,13g,0.17mol)。反应混合物在室温下搅拌过夜。反应完成后,用水稀释反应混合物,并用乙酸乙酯(EtOAc,200mL×3)进行萃取。合并有机层,用无水硫酸钠(Na2SO4)干燥,随后在减压下浓缩,得到N-(6-溴吡啶-2-基)乙酰胺(产率88%)。产物经1H NMR(400 MHz,CDCl3)表征:δ8.15(d,J = 8.0 Hz,1H),7.97(brs,1H),7.55(t,J = 8.0 Hz,1H),7.18(d,J = 8.0 Hz,1H),2.19(s,3H)。
