以吡唑和2-溴吡啶为原料合成2-吡唑-1-吡啶(Pyr-N-PzH)的一般步骤:向250 mL单颈圆底烧瓶中加入15.0 g(95 mmol)2-溴吡啶、25.0 g(367 mmol,3.86当量)吡唑和45 mL二甲苯。将反应混合物加热回流8小时,随后冷却至室温。将所得混合物溶解于二氯甲烷中,有机层用250 mL水洗涤四次(直至气相色谱法未检测到吡唑)。有机层经无水硫酸镁干燥,过滤后旋转蒸发,得到12.0 g(82.6 mmol,收率87%)目标产物,为白色固体。产物结构经1H NMR(400 MHz,CDCl3)确认:δ 8.59(m,1H,吡啶基6-H),8.39(s,1H,吡唑-5-H),7.90(d,1H,吡啶基3-H),7.80(m,1H,吡啶基4-H),7.73(s,1H,吡唑3-H),7.15(s,1H,吡啶基-5-H),6.45(m,1H,吡唑-4-H)。
