以2-氨基-5-氟苯甲酸为原料合成2-氨基-3-溴-5-氟苯甲酸的一般步骤:将N-溴代琥珀酰亚胺(631 mg,3.55 mmol)分批加入到溶解于乙酸(5 mL)中的2-氨基-5-氟苯甲酸(7,500 mg,3.22 mmol)的搅拌溶液中,反应混合物在室温下搅拌过夜。通过过滤收集所得沉淀,用石油醚(20 mL)洗涤,并干燥至恒重,得到2-氨基-3-溴-5-氟苯甲酸(11,501 mg,产率66.4%),为透明米色固体。产物无需进一步纯化即可使用。产物表征数据如下:LCMS(t R = 1.35 min,纯度= 100%),ESI m/z未检测到离子;1H NMR(DMSO-d6)δ(ppm)6.76(宽单峰,2H),7.55(双峰,J = 3.0 Hz,JH-F = 9.4 Hz,1H),7.71(双峰,J = 3.0 Hz,JH-F = 7.8 Hz,1H),12.72(宽单峰,1H);13C NMR(DMSO-d6)δ(ppm)109.4(双峰,JC-F = 7.1 Hz),111.1(双峰,JC-F = 7.1 Hz),116.2(双峰,JC-F = 22.1 Hz),124.9(双峰,JC-F = 24.8 Hz),145.1,151.4(双峰,JC-F = 235.3 Hz),168.1(双峰,JC-F = 2.7 Hz)。
