以3-氯-5-硝基甲苯为原料合成3-氯-5-甲基苯胺的一般步骤:向含有1-氯-3-甲基-5-硝基苯(5.0 g,29 mmol)的乙醇溶液(75 mL)中加入二水合氯化亚锡(SnCl2·2H2O,32.8 g,146 mmol)。将反应混合物加热回流3.0小时。反应完成后,将溶液在减压下浓缩,残余物重新溶解于氢氧化钠水溶液中,过滤,并用乙酸乙酯(EtOAc)萃取。合并有机层,用饱和食盐水洗涤,无水硫酸镁(MgSO4)干燥,减压浓缩。粗产物通过硅胶柱色谱法纯化,得到3-氯-5-甲基苯胺(4.0 g,收率97%)为浅黄色固体。产物结构经1H NMR(500 MHz,CDCl3)和ESI-MS确认:1H NMR δ 6.56(s,1H),6.48(s,1H),6.36(s,1H),3.66(bs,2H),2.23(s,3H);ESI-MS:m/z 141.7([M + H]+)。
