步骤3:5-氯-2-乙基苯胺的合成 将水合肼(6.95 mL,134.7 mmol)的甲醇(50 mL)溶液缓慢滴加至4-氯-1-乙基-2-硝基苯(6.25 g,33.7 mmol)的甲醇(120 mL)溶液中。反应体系中加入氯化铁(III)(547 mg,3.4 mmol)和活性炭(547 mg)作为催化剂。将反应混合物在回流条件下搅拌13小时。反应完成后,通过硅藻土过滤去除固体催化剂,浓缩滤液。所得粗产物通过快速色谱法(洗脱剂:己烷/乙酸乙酯 = 9:1)纯化,得到浅粉色油状目标化合物5-氯-2-乙基苯胺(5.09 g,收率97%)。 1H NMR (400 MHz, DMSO-d6) δ ppm: 1.09 (t, J = 7.51 Hz, 3H), 2.39 (q, J = 7.49 Hz, 2H), 5.13 (s, 2H), 6.47 (dd, J = 8.06, 2.20 Hz, 1H), 6.62 (d, J = 2.20 Hz, 1H), 6.89 (d, J = 8.06 Hz, 1H)。
