以3-氧代-8-氮杂双环[3.2.1]辛烷-8-羧酸叔丁酯为起始原料,合成3-羟基-8-氮杂双环[3.2.1]辛烷-8-羧酸叔丁酯的一般步骤如下:在室温下,将硼氢化钠(178 mg,4.7 mmol)缓慢加入至3-氧代-8-氮杂双环[3.2.1]辛烷-8-羧酸叔丁酯(0.50 g,2.2 mmol)的乙醇(10 mL)溶液中。反应混合物在室温下持续搅拌1小时。反应完成后,用饱和氯化铵溶液(30 mL)淬灭反应,随后用乙酸乙酯(3×20 mL)进行萃取。合并有机相,依次用水和盐水洗涤,无水硫酸钠干燥,过滤后减压浓缩,得到标题化合物3-羟基-8-氮杂双环[3.2.1]辛烷-8-羧酸叔丁酯(463 mg,收率92%),为endo和exo立体异构体的混合物。产物经GC-MS(EI)分析,分子式C12H21NO3的分子离子峰(M+)为227。
