以N-Boc-3-氮杂环庚酮为原料合成3-羟基氮杂环庚烷-1-甲酸叔丁酯的一般步骤:在0℃下,向N-Boc-3-氮杂环庚酮(5.5g,25.8mmol)的甲醇(55mL)溶液中分批加入硼氢化钠(NaBH4)(0.98g,25.8mmol),反应进程通过薄层色谱(TLC)监测。反应完成后,用盐酸(HCl)溶液将反应混合物的pH调节至5至6,随后进行过滤,并用二氯甲烷(DCM,100mL×3)萃取。合并有机相,用无水硫酸钠(Na2SO4)干燥,过滤后减压浓缩,得到3-羟基氮杂环庚烷-1-甲酸叔丁酯(油状物,4.8g,收率87%)。产物结构通过1H NMR(400MHz,d6-DMSO)确认:δppm3.95-3.93(m,1H),3.71-3.62(m,2H),3.40(m,1H),3.32-3.28(m,1H),3.03(m,1H),1.88-1.65(m,4H),1.55-1.36(m,11H)。
