以3-羰基-环丁烷甲酸甲酯为原料合成3-羟基环丁烷羧酸甲酯的一般步骤:将3-羰基-环丁烷羧酸甲酯(307.5 g,2.4 mol,1.0当量)溶解于3 L四氢呋喃中,并将溶液冷却至-78至-60℃。随后,缓慢滴加3.5 L氢化三叔丁氧基铝锂的四氢呋喃溶液(915.4 g,3.6 mol,1.5当量)。反应3小时后,通过TLC确认反应完成。然后,滴加1 L的2.5 mol/L盐酸以终止反应,并调节反应溶液的pH值至约6,同时控制温度在0℃左右。加入3 L乙酸乙酯稀释反应混合物,搅拌30分钟。通过硅藻土过滤反应混合物,滤饼用乙酸乙酯洗涤三次。分离滤液,水相用乙酸乙酯萃取。合并所有有机相,经干燥后浓缩,得到黄色液体顺式-3-羟基环丁基羧酸甲酯(273.3 g,2.1 mol),产率为88%,纯度为95%。
