步骤3:将3-氨基-4-(甲氨基)苯甲酸(280,678 mg,4.1 mmol)溶于50%甲酸水溶液中(若使用原酸酯则选用无水溶剂),于85℃搅拌反应13小时。反应完成后,将反应液浓缩,残余物重新溶解于水中,经冷冻干燥得到1-甲基-1H-苯并[d]咪唑-5-羧酸(281,712 mg,收率99%)。产物表征数据如下:1H NMR (400.2 MHz, DMSO) δ (ppm): 12.9 (br s, 1H), 8.80 (s, 1H), 8.26 (d, J = 1.6 Hz, 1H), 7.96 (dd, J = 1.6, 8.6 Hz, 1H), 7.79 (d, J = 8.6 Hz, 1H), 3.95 (s, 3H). MS: 计算值 176.2; 实测值 177.1 (M + H).
