以6,7-二氢-5H-环戊二烯并[b]吡啶1-氧化物(2.5g,18mmol)为原料,与磷酰氯(20mL,200mmol)混合。将反应混合物在120℃下搅拌3小时。反应完成后,通过减压蒸馏除去过量的POCl3。将残余物用80mL EtOAc稀释,并用Na2CO3溶液中和至pH 7-8。过滤后,水层用EtOAc萃取两次(每次40mL)。合并有机萃取液,用无水Na2SO4干燥,过滤并减压浓缩,得到4-氯-6,7-二氢-5H-环戊二烯并[b]吡啶(2.6g,收率93%)。LCMS分析:C8H9ClN(M + H)+的计算值:m/z = 154.2;实测值:154.3。
