步骤B:(i)(R)-2-甲基吡咯烷盐酸盐的合成;(i)将(R)-叔丁基2-甲基吡咯烷-1-羧酸酯(22.7g,123mmol)溶解于4M氯化氢的二恶烷溶液(100mL)中,室温下搅拌反应30分钟。通过TLC(展开剂比例为2:1的乙酸乙酯/己烷)监测反应,确认原料完全消耗。反应完成后,减压蒸馏去除溶剂。将粗产物溶于乙腈和甲苯的混合溶剂中,再次减压蒸馏去除溶剂。所得白色固体在减压条件下干燥过夜,得到标题化合物(14.6g),收率为98%。1H NMR(400MHz,CDCl3)δ1.55(d,J=6.57Hz,3H),1.66-1.77(m,1H),1.95-2.04(m,1H),2.05-2.23(m,2H),3.25-3.36(m,1H),3.37-3.48(m,1H),3.62-3.74(m,1H),9.38(s,1H),9.86(s,1H)。
