步骤A:1-甲基-1H-1,2,4-三唑-3-甲酸甲酯的制备:在氮气保护下,向0℃搅拌中的氢化钠(60%油分散体,0.346g,8.66mmol)在N,N-二甲基甲酰胺(DMF,20mL)中的悬浮液中,缓慢滴加1H-1,2,4-三唑-3-羧酸甲酯(1.00g,7.87mmol)的DMF(20mL)溶液。反应混合物在0℃下继续搅拌1小时。随后,逐滴加入碘甲烷(MeI,0.982mL,15.7mmol)。将反应混合物在室温下搅拌过夜。反应完成后,将混合物倒入冷水中,并用乙酸乙酯(EtOAc)萃取。合并有机层,用饱和食盐水洗涤,无水硫酸钠干燥,减压浓缩。残余物通过硅胶柱色谱(洗脱剂:己烷/乙酸乙酯=3:1)纯化,得到目标产物1-甲基-1H-1,2,4-三唑-3-甲酸甲酯(0.380g,收率34%),为白色固体。质谱(APCI)m/z = 142.1(M + H)。
