向4-氯丁酰氯(50.0 g,355 mmol)和N-甲氧基-N-甲基胺盐酸盐(34.6 g,355 mmol)的二氯甲烷(DCM,709 mL)溶液中,于0℃下缓慢加入三乙胺(TEA,109 mL,780 mmol),加料过程持续30分钟。反应混合物在0℃下继续搅拌3小时。反应完成后,加入水(250 mL)进行淬灭。分离有机层,用饱和盐水洗涤,无水硫酸钠(Na2SO4)干燥,过滤后减压浓缩,得到4-氯-N-甲氧基-N-甲基丁酰胺(55.0 g,产率94%)为黄色油状物,该产物无需进一步纯化即可用于后续反应。1H-NMR(CDCl3,Varian,400 MHz)δ:2.12(2H,五重峰,J = 6.4 Hz),2.63(2H,三重峰,J = 7.2 Hz),3.19(3H,单峰),3.64(2H,三重峰,J = 6.4 Hz),3.71(3H,单峰)。
