以4-溴-2-甲基吡啶(0.500 g,2.906 mmol)和联硼酸频那醇酯(0.812 g,3.997 mmol)为原料,合成2-甲基-4-吡啶硼酸频呐醇酯的一般步骤如下:在惰性气氛下,将4-溴-2-甲基吡啶、联硼酸频那醇酯、乙酸钾(0.854 g,8.720 mmol)和[1,1'-双(二苯基膦)二茂铁]二氯化钯(II)(0.021 g,0.029 mmol)溶于脱气的1,4-二恶烷(5 mL)中,置于50 mL密封管中。反应混合物在80℃下搅拌4小时。反应完成后,通过旋转蒸发除去溶剂,粗产物经硅胶柱色谱纯化(洗脱剂:乙酸乙酯/乙醚=30:70),得到2-甲基-4-吡啶硼酸频呐醇酯(0.250 g,收率39%),为灰白色固体。LCMS分析显示m/z 220.1 [M + H]+。
