以4-氯丁腈和硫脲为原料合成3-氰基丙基异硫脲盐酸盐的一般步骤:将γ-氯丁腈(50 g,48±3 mmol)和硫脲(4.04 g,53.1 mmol)溶于40 mL水中。将反应混合物加热回流3至4小时。反应完成后,蒸发除去水分,加入20 mL乙醇后再次蒸发,此操作重复三次。随后,向残余物中加入10 mL甲醇和30 mL丙酮,搅拌混合物1小时。过滤收集结晶产物,并在高真空下干燥过夜。最终得到5.44 g(30.3 mmol,收率62±7%)白色晶体产物,熔点为134-135℃,纯度大于97%。产物经1H NMR(300 MHz, d6-DMSO)确认:δ 1.89(m, 2H),2.63(t, 2H, J = 7.2 Hz),3.23(t, 2H, J = 7.2 Hz),3.38(s, 3H)。13C NMR(75 MHz, d6-DMSO)δ 15.3, 25.0, 28.8, 119.8, 169.7。分子式为C5H10ClN3S,精确质量为179.03。质谱分析显示m/z 179.03(100.0%),181.03(32.1%),180.03(7.4%),181.02(4.5%),182.03(2.0%),183.02(1.5%)。元素分析计算值(%):C 33.42, H 5.61, Cl 19.73, N 23.39, S 17.85;实测值:C 33.44, H 5.48, N 23.40, S 18.31。
