以化合物17(500 mg)为原料,将其与6M盐酸(2.2 mL)和甲醇(2.5 mL)混合,回流反应3小时。反应完成后,将混合物冷却至室温,过滤收集沉淀物。滤液经真空浓缩后,将残余物溶解于水中,用1M氢氧化钠溶液中和。中和后的溶液中析出沉淀,真空过滤收集,并依次用水和乙醚洗涤,得到目标产物5-苯基-1H-咪唑-2-胺(18,131 mg,产率33%),为浅棕色固体。产物经1H NMR(DMSO-d6)表征:δ 5.25(2H,s),6.96(1H,s),7.07(1H,t,J = 7.5 Hz),7.26(2H,t,J = 7.5 Hz),7.57(2H,d,J = 7.5 Hz),10.53(1H,br s)。质谱分析显示FAB MS m/e (M + H)+ 160;高分辨质谱(ESI)测得C9H10N3 (M + H)+的计算值为160.0875,实测值为160.0871。元素分析计算值(C9H9N3·3H2O):C,65.68;H,5.88;N,25.53;实测值:C,65.81;H,5.49;N,25.19。
