向4-(甲硫基)苯酚(50g,357mmol)、多聚甲醛(72.3g,2407mmol)和无水氯化镁(50.9g,535mmol)的乙腈(500mL)悬浮液中加入三乙胺(186mL,1337mmol)。反应混合物在室温下搅拌后,升温至60℃并持续搅拌5小时。反应完成后,将混合物冷却至0℃,缓慢加入1N盐酸直至两相分离(约1.5L)。随后加入甲基叔丁基醚(MTBE,700mL),分离有机层。有机层用饱和盐水洗涤,无水硫酸钠干燥,过滤后减压浓缩。粗产物通过硅胶快速柱色谱纯化,洗脱剂为1:1的己烷/二氯甲烷混合溶剂,得到2-羟基-5-(甲硫基)苯甲醛(50.5g,300mmol,收率84%)为半固体。
