以4-溴-3-甲氧基苯胺(102 mg,0.50 mmol)为起始原料,在0℃下将其溶于浓盐酸(10 mL)中。随后,缓慢加入亚硝酸钠(NaNO2,45 mg,0.65 mmol)的水溶液(5 mL),保持反应温度在0℃,搅拌反应1小时。接着,加入碘化钾(KI,249 mg,1.5 mmol)的水溶液(5 mL),反应混合物逐渐升温至室温并搅拌过夜。反应完成后,用乙酸乙酯萃取产物。有机相依次用水和10%硫代硫酸钠(Na2S2O3)溶液洗涤,无水硫酸镁(MgSO4)干燥,过滤后浓缩,得到2-溴-5-碘苯甲醚147 mg,收率94%。产物经质谱(ESI)分析,m/e 332(M + 20)+;核磁共振氢谱(1H NMR,300 MHz,DMSO-d6)δ 7.39(d,J = 1.87 Hz,1H),7.34(d,J = 8.11 Hz,1H),7.24(dd,J = 8.11, 1.87 Hz,1H),3.86(s,3H)。
