以2-甲基-4-(4,4,5,5-四甲基-1,3,2-二氧硼杂环戊烷-2-基)苯酚为原料合成4-羟基-3-甲基苯硼酸的一般步骤： 1. 在室温下，将高碘酸钠（4.22 g，19.74 mmol）和乙酸铵（1.52 g，19.74 mmol）加入到2-甲基-4-(4,4,5,5-四甲基-1,3,2-二氧硼杂环戊烷-2-基)苯酚（1.54 g，6.58 mmol）的丙酮和水（2:1，66 mL）混合溶液中。 2. 将反应混合物搅拌19小时，随后进行过滤并浓缩。 3. 向浓缩后的滤液中加入氯化钠，用乙酸乙酯进行萃取。 4. 合并有机层，用无水硫酸镁干燥，过滤并浓缩。 5. 通过硅胶色谱法纯化残余物，洗脱剂从1:1的乙酸乙酯:己烷逐渐过渡至纯乙酸乙酯，得到364.4 mg（产率36%）的4-羟基-3-甲基苯硼酸，为固体。 产物表征： 1H NMR (400 MHz, d6-DMSO): δ 9.31 (s, 1H), 7.55 (s, 1H), 7.52 (d, J = 8 Hz, 1H), 6.76 (d, J = 8 Hz, 1H), 2.14 (s, 3H)。 ESI-LCMS m/z 151 (M-H)-。
