以2-羟基-5-碘-3-(三氟甲基)吡啶为原料合成2-氯-5-碘-3-三氟甲基吡啶的一般步骤:在微波反应小瓶中,将5-碘-3-三氟甲基-2-吡啶醇(1g,3.47mmol)加入预先冷却的POCl3(1.60mL)和DMF(1mL)的混合液中。密封小瓶后,于110℃加热反应20分钟。反应完成后,将混合物冷却至室温,并缓慢倒入冰水中。此时,产物以沉淀形式析出。通过过滤收集沉淀,用冷水洗涤,干燥后得到浅棕色粉末状产物661mg,收率62%。产物的结构通过1H NMR(500MHz, CDCl3)δ 8.32(d, J = 2.0Hz, 1H),8.81(d, J = 2.0Hz, 1H)和13C NMR(250MHz, CDCl3)δ 89.4, 121.2(q, JC-F = 273.3Hz),126.8(q, JC-F = 33.6Hz),144.34, 148.5, 158.7进行确认。
