2-氯-5-甲氧基嘧啶-4-胺的合成步骤如下:在氩气保护下,将2,4-二氯-5-甲氧基嘧啶(10g,56.17mmol)溶于1,4-二恶烷(20mL)中,室温搅拌。随后,向该溶液中缓慢加入氢氧化铵(25.5mL)。将反应混合物转移至密封管中,加热至100℃并维持21小时。反应进程通过薄层色谱(TLC)监测。反应完成后,在减压条件下蒸除挥发性组分。粗产物通过硅胶柱色谱法纯化,洗脱剂为40%乙酸乙酯/己烷,得到2-氯-5-甲氧基嘧啶-4-胺(8.9g,收率99%),为灰白色固体。产物结构经1H-NMR(DMSO-d6, 500MHz)确认:δ7.68(s, 1H),3.81(s, 3H);LC-MS(柱:X-Bridge C-18,50×3.0mm,3.5μm;流动相:0.05%三氟乙酸水溶液/乙腈;流速:0.80mL/min)显示m/z 160([M+H]+),保留时间2.18分钟;TLC(展开剂:50%乙酸乙酯/己烷)显示Rf值为0.3。
