磷酸利美尼定性质、用途与生产工艺

用途

该药物除与α2-肾上腺素能受体结合外，与咪唑啉受体有更大的亲和力。它通过与咪唑啉受体的结合而发挥降压作用，同时也减轻了与α2-肾上腺素能受体结合带来的副作用。并且该药临床上具有耐受性好、副作用小、安全性好、疗效显著等优点。

药理作用

本品为中枢性降压药，但与可乐定不同，本品对延髓侧突喙的侧网状核的咪唑啉受体有高度亲和力，对中枢α2受体作用较弱，用药后能产生降压效果，不良反应远比可乐定轻，为第二代中枢性降压药。

制备

在装有机械搅拌、温度计、回流冷凝器的1L反应瓶中，依次加入53.0g（0.46mol）2-乙氧基噁唑啉，56.0g（0.50mol）双环丙基甲胺，200mL甲苯，充分搅拌后，升温至回流并保持回流反应8h，反应结束后，降温到25℃，向其中加入5%的碳酸氢钠溶液200mL洗涤，分层，有机相再依次用200mL饱和食盐水和200mL水洗涤，无水硫酸钠干燥，过滤，浓缩得76.2g粗品，粗品再用环己烷重结晶，得75.0g白色固体，含量99.7%，m.p.105.5~107℃，收率90.5%。利美尼定与磷酸成盐制备磷酸利美尼定。

生物活性

Rilmenidine Phosphate 是选择性的I(1) imidazoline receptor激动剂,用于治疗高血压。

体外研究

Rilmenidine provides antihypertensive efficacy comparable with that of diuretics, beta-blockers, calcium channel blockers, and angiotensin-converting enzyme (ACE) inhibitors.
Rilmenidine phosphate (25-100 μM; 24 hours) inhibits K562 cell proliferation.

Cell Proliferation Assay

Cell Line: K562 cells
Concentration: 25, 50, 100 μM
Incubation Time: 24 hours
Result: Dose-dependently inhibited K562 colony formation.

体内研究

Rilmenidine phosphate-treated N171-82Q mice (i.p.; 4-times a week) displays significant improved forelimb grip strength and all limbs grip strength from 12 to 22 weeks of age.
Rilmenidine phosphate decreases levels of mutant huntingtin.
