1,3-二溴-5-氯苯性质、用途与生产工艺

应用

1，3-二溴-5-氯苯是一种重要的化工原料和药物中间体，是制备口服生物可利用的1-（1H-吲哚-4-基）-3，5-二取代的苯类似物作为抗有丝分裂剂的反应物。

制备方法

将1，3，5-三溴苯（9.44g，30mmol）在120mL乙醚中的溶液在干冰/丙酮浴中冷却至-78℃。在10分钟内逐滴加入正丁基锂（13.2mL的2.5M己烷溶液，33mmol）。将所得混合物在-78℃下搅拌另外10分钟，然后在3分钟内分小份加入六氯乙烷（7.15g，30.2mmol）。然后将反应混合物在-78℃下搅拌15分钟，然后在室温下搅拌3.2小时。 将混合物在100mL水和100mL EtOAc之间分配。 分离水层并用另外的100mL EtOAc萃取。 然后将合并的有机层用MgSO 4干燥，过滤，真空浓缩，得到1，3-二溴-5-氯苯，为浅棕色固体（7.72g，95％）。
