头孢氨苄EP杂质C性质、用途与生产工艺
头孢氨苄为半合成的第一代口服头孢菌素,是对革兰阳性、阴性细菌都有杀菌作用的广谱抗生素。临床适用于泌尿道、呼吸道、皮肤和软组织、生殖器官(包括前列腺)等部位的感染,也常用于中耳炎。头孢氨苄EP杂质C是头孢氨苄重要的杂质。
将N-BOC-邻苯基甘氨酸(753mg,3mmol)溶解在四氢呋喃(THF)(15ml),Et3N0.42ml(3mmol)和3滴N-甲基吗啉的混合物中,在-15℃下搅拌10分钟。逐滴加入在2mlTHF中的氯甲酸异丁酯(0.41ml,3mmol)。在-15°C搅拌20分钟后,将1.1g(3mmol)头孢氨苄溶解在10ml含0.42in1(3mmol)Et,N的THF-H2O(1:l)中。加入。将混合物在0℃下搅拌3小时。通过蒸发除去THF。加入H2O(10ml),并用2NHCl将pH调节至2。然后将溶液用EtOAc(3×20ml)萃取。合并的组织。将水层以H2O(20毫升)洗涤,以无水MgSO4干燥,过滤并蒸发。将残余物溶解在约2ml的氟乙酸中,并在室温下保存。持续16小时。蒸发溶液,将溶于水的残余物置于阴离子交换柱(Bio-RadAG-3,200-400目,乙酸盐,13×2cm)上,并用水洗脱以除去三氟乙酸。蒸发洗脱液,残余物从丙酮-水中结晶并真空干燥。头孢氨苄EP杂质C产率900mg(63%)。
