4-METHYL-2-(1-PIPERIDINYL)-QUINOLINE性质、用途与生产工艺
ML204是新型的、有效的、选择性的TRPC4抑制剂。在胞内钙离子荧光分析试验中,IC50为1 μM。在全细胞电压钳实验中,IC50约为3 μM。ML204对TRPC通道的选择性远高于对其他TRP通道和非TRP通道的选择性。
ML204抑制TRPC4β介导的胞内钙离子上升,IC50为0.96 μM,其选择性是对毒蕈碱型受体耦合的TPRC6通道的19倍。ML204对TPRC4β的抑制不依赖于GPCR的激活。选择性实验表明,10-20 μM ML204对TRPV1, TRPV3, TRPA1, TRPM8和KCNQ2和小鼠背根神经节神经元中的电压依赖性钠离子通道、钾离子通道和钙离子通道没有明显抑制作用。ML204可抑制由G
