利索茶碱性质、用途与生产工艺
利索茶碱可用作医药合成中间体。利索茶碱的合成可使用钴类化合物作为催化剂。钴作为地球富含的元素,廉价易得,并且它是维生素B12的重要组成元素,在生命体内广泛存在,生物毒性相对较小。
在无水无氧手套箱中,将钴络合物(0.005~0.04mmol,0.5~4mol%)或者原位卡宾钴催化体系[即CoCl2(0.005~0.04mmol,0.5~4mol%)和式3氮杂环卡宾配体(0.005~0.04mmol,0.5~4mol%)的混合物]、以及相应当量的碱(0.05~0.2mmol,5~20mol%)加入到含有磁力搅拌子的4mL大小玻璃反应瓶中,随后加入0.5mL的溶剂,最后加入底物(1mmol),反应摇晃30秒后装入ParrInstruments高压反应釜。将高压反应釜移出手套箱后通入10~30bar氢气,将反应温度设定在25~60℃反应16小时。反应停止后,高压反应釜先置于零度冰水浴中冷却,然后缓慢释放出釜内氢气。将反应瓶从高压反应釜中取出,然后使用快速柱层析将产物利索茶碱分离并称量得到产率。
