钙调神经磷酸酶底物性质、用途与生产工艺
钙调神经磷酸酶是哺乳动物脑内含量极丰富的唯一依赖Ca2+及钙调素的磷蛋白磷酸酶。该酶(calcineurin,CaN)在脑外组织如精子细胞、淋巴细胞及肌肉组织中也有分布,但含量远远低于脑。CaN由18kDCa2+结合的调节亚基和61kD钙调素结合的催化亚基组成。除可与Ca2+结合外,该酶还可与Mn2+,Ni2+,Co2+等金属离子结合而影响其活性。
免疫组织化学定位表明,它主要分布在大脑纹状体、海马、大脑皮层等区域,在各个脑区CaN的多种同工酶在特定神经元亚群有不同表达,CaN这种区域性表达可能与不同钙调神经磷酸酶底物专一性有关。
Calcineurin substrate 是来自 cAMP 依赖性蛋白激酶调节性 RII 亚基的多肽。Calcineurin substrate 可用于钙调神经磷酸酶活力的测定。
Calcineurin (PP2B), a serine/threonine phosphatase controlled by cellular calcium, has been implicated in a wide variety of biological responses including lymphocyte activation, neuronal and muscle development, neurite outgrowth, and morphogenesis of vertebrate heart valves.
