定義
チューブリンは，微小管の主要構成成分をなすタンパク質。単量体はそれぞれ分子量約5万5000のα(アルファ)とβ(ベータ)で、ともにその全アミノ酸配列はブタの脳から得られたものについて決定されている。二量体一に2分子のグアニンヌクレオチドが、一つは強く、他は弱く結合している。アルカロイドのコルヒチン、ビンクリスチン、ビンブラスチンが強く結合し、微小管の重合を阻害する。これにより有糸分裂阻害がおこる。分子内にグアニンヌクレオチドを含み，条件によって重合または脱重合を起こす。微小管を形成して細胞骨格やさまざまな細胞運動（繊毛運動や有糸分裂など）に関わるほか，神経の軸索流など細胞内の物質輸送にも関与，細胞生理に重要な役割をする多機能のタンパク質である。

解説
微小管microtubuleを構成するタンパク質。微小管は生物界のほとんどすべての細胞に見いだされる直径24nmの管状繊維構造で，鞭毛･繊毛運動，細胞形態の形成･維持，細胞分裂，神経軸索内輸送などの多くの生理現象に深く関与している。1963年にギボンズI.R.Gibbonsが抽出，68年に毛利秀雄が命名。チューブリンはα,βの2種の異なったサブユニットから成り，異質二量体としての分子量は約11万で，2分子のGTP(グアノシン三リン酸)と1分子のマグネシウムを結合している。

The protein subunit of microtubules in eukaryotic cells. Tubulin exists in the
cell as a dimer of α- and β-tubulin, each with a molecular
weight of about 55,000 daltons as determined by electrophoresis. α-Tubulin contains 450 amino acids, with a very acidic Cterminal sequence. β-Tubulin has 445 amino acids with about
42 percent sequence identity with the α-subunit, which suggests a common ancestral origin. The tubulin dimer is associated with two moles of ATP, one at an exchangeable site and
the other at a non-exchangeable site. Under appropriate conditions of temperature, pressure, ionic strength and calcium concentration, tubulin dimers self-assemble to form the cytoplasm
microtubule complex of interphase cells and the spindle of
dividing cells.
