定義

4-Phenylbutyric acid is a monocarboxylic acid the structure of which is that of butyric acid substituted with a phenyl group at C-4. It is a histone deacetylase inhibitor that displays anticancer activity. It inhibits cell proliferation, invasion and migration and induces apopto is in glioma cells. It also inhibits protein isoprenylation, depletes plasma glutamine, increases production of foetal haemoglobin through transcriptional activation of the gamma-globin gene and affects hPPARgamma activation.

酵素阻害剤

This short-chain fatty acid and ammonia-scavenging pro-drug (FWfree-acid = 164.20 g/mol; CAS 1821-12-1; Abbreviation: PBA), which is administered orally as sodium phenylbutyrate (FWsodium-salt = 186.18 g/mol; CAS 1716- 12-7; Trade Name = Buphenyl®; Abbreviation: NaPBA), is an FDA- approved treatment for hyperammonia as a result of urea cycle disorders. Primary Mode of Action: PBA undergoes two-carbon loss to form the active drug, phenylacetate, which is then enzymatically conjugated to glutamine to form phenylacetylglutamine, with the latter excreted in urine. Because glutamine contains two equivalents of ammonia, PBA is especially effective in its action. Twenty-four-hour urinary phenylacetylglutamine levels correlate strongly with dose, making it a clinically useful non-invasive biomarker for compliance and therapeutic monitoring. Glycerol phenylbutyrate (FW = 530.67 g/mol; CAS; 611168-24-2; IUPAC Name: propane-1,2,3-triyl-tris (4-phenylbutanoate; Other Names: HPN-100 and Ravicti®) is a non-sodium-releasing FDA-approved pro-pro-drug that releases PBA and glycerol upon metabolic hydrolysis. Other PBA Actions: Sodium PBA also acts as a chaperone, reducing the load of mutant or unfolded proteins retained in the ER during cellular stress. For example, PBA protects against liver ischemia re-perfusion injury by inhibiting endoplasmic reticulum stress-mediated apoptosis. By stimulating fetal hemoglobin synthesis, PBA is useful in treating sickle cell disease. PBA also induces Arginase I in many tissues (by as much as 4x in brain), a finding that suggests reduced arginine may be manifested as reduced nitric oxide synthesis. Historical Note: In his classic studies elucidating the b- oxidation pathway, Knoop used phenylbutyrate to demonstrate that, after oral administration, phenylacetylglycine (phenaceturic acid) appears in the urine. This observation suggested phenylbutyrate underwent one cycle of two-carbon chain shortening, forming phenylacetate, which was then coupled to glycine.
