硫酸プロタミン 化学特性,用途語,生産方法
解説
硫酸プロタミン，血液凝固作用をもつヘパリンの拮抗(きっこう)剤で、サケ科などの魚類の成熟精巣から抽出したプロタミンの硫酸塩をいう。分子量1万以下の強塩基性単純タンパク質で、熱では凝固しない。核酸やヘパリン、インスリンのような高分子化合物と結合して沈殿を生ずる。
用途
ヘパリンの過量投与の際、中和の目的で投与される。また、血液透析や人工心肺など血液体外循環のヘパリン作用の中和にも利用される。そのほか、インスリンと結合させ、インスリンの作用時間を延長させる製剤プロタミンインスリン亜鉛水性懸濁注射液として糖尿病の治療に用いられている。製剤としては、硫酸プロタミン注射液（10ミリリットル中100ミリグラム）が市販されている。なお、プロタミンそのものも血液凝固阻止作用があり、止血剤として手術前後の出血防止にも用いられる。

Protamine sulfate has an anticoagulanteffect, but if used in the proper amount, it counteractsthe action of heparin and is used as an antidote for thelatter in cases of overdosage. It is administered intravenouslyin a dose that depends on the circumstances.
