물성

백색∼담황색의 결정 또는 결정성 분말로 냄새는 없고 에탄올-아세톤 혼합액으로 재결정하면 판상 결정 또는 기둥상 결정이 얻어진다. 물에는 용해되기 쉽고(19.4g/100g-H2O 25℃), 알코올에는 25℃에서 0.47g/100g, 아세톤, 에테르, 클로로포름에는 용해되지 않는다. 열, 산, 알칼리에 안정하나 중성, 알칼리성에서는 광선에 약하고 산화제에도 약하다. 중성용액에서는 120℃에서 중합이 일어나는데 산성, 알칼리성에서는 중합이 일어나지 않고 안정하다.

개요

비타민B6는 동·식물계에 널리 분포하고 있으며 특히 곡류, 녹색식물, 맥주효모, 계란 노른자위, 간, 신장에도 많다. 이는 단백질 대사에 필요한 인자로 작용하고, 이의 결핍은 피부염, 습진, 빈혈 등을 일으킨다. 사람에게서 하루의 필요량은 1∼2㎎이다.

용도

주요 용도로는 강화제, 연구개발(시약용) 등에 사용한다.

순도시험

  (1) 융점 : 이 품목의 융점은 203～209℃이어야 한다.

  (2) 액성 : 이 품목의 수용액(0.5→25)의 pH는 2.5～3.5이다.

  (3) 납 : 이 품목 5.0g을 취하여 원자흡광광도법 또는 유도결합플라즈마발광광도법에 따라 시험할 때, 그 양은 2.0ppm 이하이어야 한다.

  (4) 염화물 : 이 품목 0.5g을 메탄올 50mL에 녹이고, 빙초산 5mL 및 에오신Y시액 2～3방울을 가한 다음, 0.1N 질산은용액으로 적정할 때, 그 양은 건조물로 환산하여 16.9～17.6% 이어야 한다.

0.1N 질산은용액 1mL  = 3.545mg Cl

    에오신Y시액 : 에오신Y 50mg을 물 10mL에 녹인다.

확인시험

  (1) 이 품목의 수용액(1→10,000) 1mL에 2,6-디브롬퀴논클로로이미드무수알콜용액(1→4,000) 2mL 및 암모니아시액 1방울을 가하면 청색을 나타낸다. 또한 미리 붕산포화용액 1mL를 가한 다음 이 시험을 하면 청색을 나타내지 아니한다.

  (2) 이 품목은 확인시험법 중 염화물반응을 나타낸다.

정량법

  이 품목 약 0.4g을 정밀히 달아 빙초산(비수적정용) 60mL 및 초산제이수은시액(비수적정용) 10mL를 가하여 녹인 다음 0.1N 과염소산용액으로 적정한다(지시약 : 크리스탈바이올렛․빙초산시액 1mL). 따로 같은 방법으로 공시험을 한다.

0.1N 과염소산용액 1mL ＝ 20.56mg C₈H₁₁O₃N•HCl

강열잔류물

  이 품목의 강열잔류물은 0.1% 이하이어야 한다.

Pyridoxine hydrochloride has been used:
As a vitamin component in a chemically defined diet with balanced metal content for metal depletion studies;
As a component of chemically defined medium (CDM) to culture Staphylococcus aureus WSPPA;
As analytical reference standards for the determination of analytes in pharmaceutical preparations by chromatography and spectrophotometric techniques;
As a vitamin to study the impact of protein expression and post-translational modifications specifically charge heterogeneity;
As a medicine, it is used to prevent or treat certain neurological disorders (peripheral neuropathy) caused by certain drugs (e.g. isoniazid). It is also used in the treatment of certain hereditary disorders (e.g. flavoururic aciduria, hyperoxaluria, homocystinuria).

일반 설명

Vitamin B6 has six forms namely, pyridoxal, pyridoxine, pyridoxamine, pyridoxamine 5′-phosphate, pyridoxine 5′-phosphate and pyridoxal 5′-phosphate. Pyridoxine is a is a water-soluble vitamin and a pyrimidine derivative of vitamin B6. It is considered heat and acid stable.

Safety Profile

Poison by intravenous route. Moderately toxic by ingestion, intramuscular, and subcutaneous routes. Human reproductive effects by ingestion and intramuscular routes: postpartum changes. Experimental reproductive effects. Human mutation data reported. When heated to decomposition it emits very toxic fumes of NOx and HCl.

Purification Methods

Purify the vitamin by recrystallisation from EtOH/Me2CO, n-BuOH or MeOH/Et2O. Its solubility in H2O is 22%, and in EtOH it is 1.1%. It is insoluble in Et2O and CHCl3. Acidic aqueous solutions are stable at 120o/30minute. The free base has m 159-160o after recrystallisation from Me2CO and sublimation at 140-145o/0.0001mm.
