The Notch-regulated ankyrin repeat protein (NRARP) is a small, evolutionarily conserved protein that functions as a key modulator of the Notch signaling pathway. Notch signaling plays critical roles in cell fate determination, tissue development, and homeostasis, with dysregulation linked to cancers and developmental disorders. NRARP contains two ankyrin repeat domains that mediate protein-protein interactions, enabling its role in fine-tuning Notch activity. It acts as a negative feedback regulator by promoting the degradation of Notch intracellular domain (NICD), thereby attenuating Notch target gene expression. Additionally, NRARP interacts with Wnt signaling components, suggesting crosstalk between Notch and Wnt pathways.

NRARP antibodies are essential tools for studying its expression, localization, and function in various biological contexts. These antibodies enable detection of endogenous NRARP protein in techniques like Western blotting, immunohistochemistry, and immunofluorescence. Researchers use them to investigate NRARP's role in developmental processes (e.g., cardiovascular and lymphatic system formation) and diseases such as T-cell leukemia, where Notch signaling is frequently hyperactivated. Specific NRARP antibodies also help elucidate post-translational modifications and protein interaction networks. Validated antibodies with high specificity are critical, as NRARP's small size (~25 kDa) and structural homology with other ankyrin repeat proteins pose challenges for accurate detection.
