DP1 antibody targets the prostaglandin D2 receptor 1 (PTGDR, also known as DP1), a G protein-coupled receptor that binds prostaglandin D2 (PGD2), a lipid mediator involved in inflammation and immune regulation. Discovered in the 1980s, PGD2 is primarily produced by mast cells, Th2 cells, and dendritic cells, playing a dual role in both promoting and resolving inflammatory responses. DP1 receptor activation triggers cAMP signaling, influencing smooth muscle relaxation, vasodilation, and immune cell recruitment. Dysregulation of the PGD2-DP1 axis is linked to allergic diseases (e.g., asthma, allergic rhinitis), chronic inflammation, and tumor microenvironment modulation.

DP1 antibodies are essential research tools for studying receptor localization, expression patterns, and signaling mechanisms. Therapeutically, DP1 antagonists or blocking antibodies have been explored to inhibit excessive PGD2 activity in allergies. However, clinical translation remains challenging due to the receptor's complex roles in homeostasis. Recent studies also highlight DP1's involvement in neuroinflammation and sleep regulation, broadening its potential applications. Current research focuses on developing isoform-specific antibodies and optimizing pharmacokinetics for targeted therapies. DP1 antibodies thus represent a bridge between understanding PGD2 biology and creating precision treatments for immune-related disorders.
