Hsc70 (Heat Shock Cognate 70) is a constitutively expressed member of the HSP70 (Heat Shock Protein 70) family, sharing structural and functional similarities with inducible HSP70 but differing in its baseline expression under normal cellular conditions. It plays a critical role in protein homeostasis, assisting in folding nascent polypeptides, facilitating protein translocation across membranes, and promoting the refolding or degradation of misfolded proteins via chaperone-mediated autophagy. Hsc70 also participates in clathrin-coated vesicle disassembly and antigen presentation in immune cells.

Antibodies targeting Hsc70 are widely used to study its expression, localization, and interactions in diverse biological contexts. Due to its ubiquitous presence, Hsc70 serves as a marker for cellular stress responses and proteostasis mechanisms. Researchers employ these antibodies in techniques like Western blotting, immunohistochemistry, and immunoprecipitation to investigate its roles in neurodegenerative diseases (e.g., Alzheimer’s, Parkinson’s), cancer (where Hsc70 may modulate apoptosis or drug resistance), and viral infections (as some pathogens hijack Hsc70 for replication).

Specificity is crucial when using Hsc70 antibodies, as the protein shares ~85% sequence identity with inducible HSP70. High-quality antibodies are validated to avoid cross-reactivity, often through knockout controls or peptide blocking assays. Commercial Hsc70 antibodies are typically raised against unique epitopes in its C-terminal domain.
