背景介绍
LRRC15（富含亮氨酸重复序列蛋白15）亦称为LIB和hLib。该蛋白在多种实体瘤中高表达，见于众多实体瘤（如乳腺癌、头颈癌、肺癌、胰腺癌）的基质成纤维细胞表面，并直接表达于部分间充质来源的癌细胞（如肉瘤、黑色素瘤、胶质母细胞瘤）上。TGFβ可诱导活化成纤维细胞（αSMA+标志）和间充质干细胞表达LRRC15。这些发现共同表明，LRRC15可作为新型癌症相关成纤维细胞（CAF）和间充质标志物，为治疗LRRC15阳性间质纤维化或间充质来源的癌症提供了潜在治疗靶点。ABBV-085是一种靶向LRRC15的抗体药物偶联物（ADC），含单甲基澳瑞他汀E（MMAE）有效载荷。临床前研究显示，作为单药或与标准疗法联用时，该药物对LRRC15基质阳性/癌细胞阴性模型及LRRC15癌细胞阳性模型均具有显著抗肿瘤活性。
