背景介绍
FAP（亦称seprase）是一种II型跨膜丝氨酸蛋白酶。其质膜形式与可溶形式均具有脯氨酸后切割内肽酶活性，尤其倾向于识别Ala/Ser-Gly-Pro-Ser/Asn/Ala共识序列。该酶还能降解明胶和热变性I型胶原蛋白，同时具备二肽基肽酶活性，优先作用于Ala-Pro、Ile-Pro、Gly-Pro、Arg-Pro及Pro-Pro等二肽序列。质膜形式的FAP通过与DPP4、PLAUR或整联蛋白结合，参与细胞外基质（ECM）的周细胞蛋白水解过程，从而促进细胞在ECM中的粘附、迁移和侵袭。该酶通过降解蛋白聚糖brevican促进胶质瘤细胞在脑实质中的侵袭。在黑色素细胞中，FAP通过以不依赖丝氨酸蛋白酶活性的方式调控细胞增殖与存活，发挥肿瘤抑制因子作用。
